How environment constrains metabolism in cancer

Abstract: Changes in cell metabolism are necessary to support cancer, and accumulating evidence suggests that cancer metabolic adaptations can also promote cancer progression. How specific cancers use metabolism to support proliferation is influenced by factors in the tumor microenvironment, including nutrient availability, with nutrient availability in tissues differing from those found in blood. We have found that altered levels of nutrients present in different environmental contexts can influence the tissues where cancer can grow. Furthermore, the metabolic constraints imposed by the nutrient environment can impact factors that promote cancer progression. Using a combination of genetic manipulations and mouse models, together with culture in different nutrient environments, data on how nutrient cooperate to constrain proliferation in different environments will be discussed.
